چکیده

Background Behçet’s disease (BD) is a chronic systemic vasculitis characterized by the overactivation of neutrophils and macrophages. Exosomes are membrane-derived vesicles that mediate intercellular communications neutrophil-derived exosomes account for major portion serum in BD. However, role BD remains unknown. Objectives 1) To investigate production neutrophils; 2) elucidate regulation macrophage exosomes; 3) explore mechanism immunoregulatory functions exosomes. Methods healthy control (HC) were extracted quantified. Human monocyte-derived macrophages (HMDM) stimulated with HC exosomes, TNF-α IL-6 examined. Differently expressed miRNAs analyzed using miRNA sequencing. LPS-induced HMDM treated mimics or inhibitors, detected. was overexpressed macrophages, RNA sequencing performed to analyze regulating pathways. Dual-luciferase assays confirm miRNA-mRNA interaction. Results produced significantly lower level than ones. Both suppressed but lesser extent Six downregulated presented including miR-122-5p. miR-122-5p inhibited while inhibitor promoted HMDMs. Overexpression attenuated TLR4 IFN-β signaling. directly targeted 3’UTR IRF5, TF pathway autocrine IFN-β, IRF5 expression confirmed dual luciferase assay. Knocking down dampened Figure 1. (A) Decreased (B) Reduced suppression activation (C) Differentially (downregulated) (D) JAK-STAT signaling HMDM. (E) (F) Conclusion Our findings suggested reduced function mediated levels Impaired might be implicated References [1]Kolonics, Ferenc et al. MiR-490-5p Functions as an OncomiR in Breast Cancer by Targeting NFATc4

Breast cancer is a serious health problem worldwide in women. MicroRNAs are small non-coding RNAs of 18–25 nucleotides in length that post-transcriptionally modulate gene expression. MiR-490 has been reported as a tumor suppressor and oncomiR microRNA in breast cancer with two separate targets, NFAT and Rho. NFAT is one of the targets for miR-490 but the relationship between hsa</e...

Decreased miR-155-5p, miR-15a, and miR-186 Expression in Gastric Cancer Is Associated with Advanced Tumor Grade and Metastasis

Background: Gastric cancer (GC) is one of the most prevalent cancers with a high rate of mortality in the world. In recent years, microRNAs (miRNAs) have been proposed to be involved in GC development. In this study, we aimed at investigating differential expression level of miR-155-5p, miR-15a, miR-15b, and miR-186 in GC. Methods: For this research, we used qPCR to investigate miR-15b, miR-155...

Exosomes derived from miR-122-modified adipose tissue-derived MSCs increase chemosensitivity of hepatocellular carcinoma

BACKGROUND Hepatocellular carcinoma (HCC) displays high resistance to conventional chemotherapy. Considering that microRNA-122 (miR-122) performs an essential function to promote chemosensitivity of HCC cells, an effective vehicle-mediated miR-122 delivery may represent a promising strategy for HCC chemotherapy. An increasing interest is focused on the use of exosomes as biological vehicles for...

miR-122 Regulates Tumorigenesis in Hepatocellular Carcinoma by Targeting AKT3

MicroRNAs (miRNAs) have been implicated in the orchestration of diverse cellular processes including differentiation, proliferation, and apoptosis and are believed to play pivotal roles as oncogenes and tumor suppressors. miR-122, a liver specific miRNA, is significantly down-regulated in most hepatocellular carcinomas (HCCs) but its role in tumorigenesis remains poorly understood. Here we iden...

Differential expression of miR-1297, miR-3191-5p, miR-4435, and miR-4465 in malignant and benign breast tumors

Objective(s): MicroRNAs (miRs) are a class of small non-coding RNAs which are associated with tumor growth and progression. In the present study, we assessed the expression of selected miRs in malignant, benign, and adjacent normal breast tissues. Materials and Methods: The expression of miR-1297, miR-3191-5P, miR-4435, and miR-4465 were...
